ACTIVITY DEPARTMENT:

  • Study of the regularities of molecular mechanisms of virus-cell interaction on the model of human adenovirus, Epstein-Barr virus, herpes simplex virus type 1 and type 2.
  • Determination of new substances with potential anti-herpetic and anti-adenoviral properties and establishing of targets of their action, that will allow to perform their structural and functional analysis with subsequent correction of direction of synthesis to create highly effective antiviral agents.
  • Development of a strategy for the investigation of antiviral activity of substances of various nature in vitro and in vivo systems; carrying out of preclinical studies.
  • Development of biotechnological aspects of the creation of specific immunobiological drugs directed action and diagnostic tools.

OUR ACHIEVEMENTS:

The study of human and animal viruses at the Institute of microbiology and virology of the National academy of sciences of Ukraine was started in 1963 in the department of biophysics of viruses (Y.G. Kishko). In 1974, the department of biophysics of viruses was reorganized into the laboratory of animal viruses, which in 1978 was renamed the department of molecular biology of viruses under the leadership of corresponding member of the National academy of sciences of Ukraine, doctors of biological sciences, Dyachenko Nataliya Sergiivna. 2004, the department has been renamed into the laboratory of viruses reproduction, which was headed until 2014 the candidate of biological sciences Nesterova Nadiya Vitaliivna. Since 2016, the laboratory of viruses reproduction has been reorganized into the department of viruses reproduction under the leadership of the candidate of biological sciences Zagorodnya Svitlana Dmytrivna.

The perennial scientific achievements of the department's staff are:

  • an in-depth study of the physico-chemical properties of human adenovirus proteins, which made it possible to obtain new fundamental knowledge about the peculiarities of the localization of adenoviral proteins in the infected cell and to discover several new antigenic determinants of the hexon and the appendage of adenovirus;
  • the different nature of polypeptides, which depends on the spatial organization of proteins, and the localization of some epitopes with the help of chemically synthesized antigenically active peptides, which simulate them, are revealed;
  • significant regularities in the structural and functional organization of adenoviruses hexon based on a comparative analysis of the antigenic specificity and the primary structure of proteins of different types of adenoviruses, which are taxonomically remote from each other, are determined;
  • the concept of immuno-inactivation (neutralization of infectiousness) of adenoviruses has been developed and for the first time a mathematical model of this process was created, which determines the contribution of antibodies to several antigenic determinants of hexone and appendage, as well as interferon and complement.

Directed search and study the mechanisms of action anti adenovirus substances in the department started in 1979 in conjunction with the doctor of chemistry Chernetsky V.P. and his learners Ph.D. Alexeyeva I.V. and Shalamay A.S. (Institute of Molecular Biology and Genetics of the National Academy of Sciences of Ukraine). The effect of the original structural analogue of cytidine (2-β-D-ribofuranosyl-5-amino-1,2,4-triazine-3 (2H) -OH) -6-azacytidine on the reproduction of the human adenoviruses has been studied the most detailed. The drug was patented as an antiviral, with low cytotoxicity and high anti-adenovirus affect. It is established that its antiviral activity is associated with suppression of the synthesis of viral DNA and proteins. A number of 6-azacitidine derivatives have been studied and compounds with less toxicity and better solubility have been determined. Anti-adenovirus action was found in 6-azauridin, a structural analogue of uridine synthesized by Ukrainian chemists. The activity of the cytosine arabinoside-cyclocytidine derivative and structural analogue of ribavirin-ribamidil, synthesized by Corresponding Member of the Latvian Academy of Sciences M.Yu. Lidak. It has been shown that ribamidyl affects at the formation of a functionally complete hexone and the maturation of adenovirion. The antiviral activity of proteolysis inhibitors ambene and ε-aminocaproic acid was studied. The target of their action is a violation of the morphogenesis of mature infectious adenovirions, caused by a violation of the processing of the internal polypeptide (VII) predecessor. Together with the staff of the A.V. Bogatsky Physico-Chemical Institute of the National Academy of Sciences of Ukraine (doctor od chemistry V.E Kuzmin, Ph.D. A.G.Artemenko) the antiviral properties of macrocyclic pyridinophanes were investigated using 3D and 4D QSAR modeling, which allows to establish the relationship between the structure and activity of a substance, and also to predict the effectiveness of compounds with respect to viruses. Using QSAR and simplex representation, active structures were found and new nitrogen-containing analogs were synthesized for which encouraging results of antiviral action were obtained. The work was carried out within the framework of grant INTAS 94-292. Antidenoviral activity of some substances isolated from plants and marine organisms is shown. The most active was Giporamin from sea-buckthorn, obtained from the RPA "VILAR" (Russia). Hyporamine blocks the reproduction of adenovirus at the stage of penetration and stripping of the virus. Water extracts from the cysts of Artemis were obtained by Dr. B. I. Rudnev and Dr. V.G.Shaida from the Institute of Southern Seas of the National Academy of Sciences of Ukraine and were patented as antiviral agents with anti-adenoviral and anti-herpetic activity.

The first domestic immunoassay test system for the diagnosis of Epstein-Barr virus-induced diseases was developed. The antigen has been obtained, its specificity proved, panels of positive and negative sera have been created. The conditions for carrying out of the immunoassay analysis were optimized and the parameters of specificity, sensitivity and compliance of the ”IFA-AtVEB-strip” test system were characterized.

In the framework of the INTAS 011-2382 grant, the effect of the Epstein-Barr virus on signaling processes in malignant B-cells transformed by the virus has been investigated. Experimental systems of virus-cells have been developed, in which the influence of viral proteins on the process of the drug induced CD-95 mediated apoptosis was investigated. Data on the influence of different levels of viral infection to the sensitivity of the cells BL-41 and DG-75 to the Etoposide and Doxorubicin, which are anti-tumor agents, were obtained. It may be due to different mechanisms of their action, which in one way or another limit the EBV replication mechanism.

In the framework of the National space program of Ukraine (2003-2007), research was carried out on the effects of simulated microgravity conditions on adenovirus and on the cell-virus system. It has been established that intact human adenovirus is able to keep infectious for a long time and reproduce in the conditions of modeled gravity (Space Science and Technology, 2003, 2004 and 2007).

 Studies, performed in the department in recent years, have been devoted to the modeling of mixed adeno-herpetic infection on the populations of the human and animal cells, and further study the effects of antiviral drugs at the developed models. The studies continues the long-term scientific researches of the collective and harmoniously combine with the direction of the development of world virology, in particular with regard to adeno- and herpesvirus. For the first time, model systems of co-infected cell cultures have been obtained, a virus replication has been reproduced in them, and a detailed analysis of structural-functional virus-cell interaction has been carried out. The anomalous action of the antiviral substances was detected by us, under the condition of co-infection of cells with two viruses, indicates the necessity of using such methodical approaches to the study of promising antiviral substances. The proposed approach of research of promising antiviral compounds on the model of mixed viral infection can take place at the creation of new and studying the activity of existing chemotherapeutic drugs, that may help to prevent the development of virus resistance to drugs. The department employees pay great attention to the urgent issue of combining scientific achievements with the practical needs of today, in particular in medicine. The methodical recommendations for preclinical studies of specific antiviral action of medicinal products in cell culture on the model of adenoviruses have been developed. The use of the proposed unified methods will contribute not only to the introduction of highly effective means into medical practice, but also to the exit of domestic drugs on the world market (Shcherbinskaya A.M., Dyachenko N.S., Rybalko S.L., Nosach L.N., Dyadyun S. Pre-clinical research of a specific antiviral action of drugs in cell culture on the model of adenovirus. Guidelines // Journal of Pharmacology and Pharmacy. - 2007, №9.- P. 52-64). Together with "BIOPHARMA PLASMA", specific immunoglobulins against TORCH infections (cytomegalovirus, herpes 1, 2 and 6 types, Epstein-Barr virus and Chlamydia) have been developed for the production. These drugs are successfully used in the treatment of the above-mentioned infections in medical institutions of Ukraine (in urological, obstetric and neurological departments).

For diagnostic purposes, immunosensors have been developed to detect specific antibodies to Epstein-Barr virus, adenovirus and herpes simplex virus in blood serum of patients using optoelectronic device "Plasmon-6".

Scientific achievements of employees were repeatedly marked with awards and scholarships, in particular, the Award "Intellectual Potential of Young Scientists - the City of Kyiv" (Zagorodnya SD, 2003); Prize to them D.K.
